plan: - (morning) talk to Zahid about - installation of the translation stage - using Psaltis' machine shop - drill two holes for the current setup - (afternoon) - continue imaging - (evening) - AFM - ssRNA 1D assembly (9.27 am) - waiting for Zahid, browsing Nikon's microscopy U website (10 am) - make the tubes co-axial - drills two holes for 8-32 screws - cheat: add sticky notes to center the black box to shift the black box forward (12 pm) - project the beam using a blank sample (MQ H_2O) on the room ceiling --> not much different from the previous alignment (1 pm) - lunch (4.54 pm) - sample: 1/10x 1um bead (95 uL MQ of H_2O and 10 uL of stock solution of bead) could not get any image --> not sure whether the new alignment made it better or worse (5.36 pm) - switched the upper and lower filters - upper filter : (550LP 7513) - lower filter : (HQ575/50 M 54652) --> the image looks much better, why? - some bad news - some of the bead stuck on surface - the number of beads in the field of view is much less than expected (6.11 pm) tommorow - ask the post-doc about Psaltis' machine shop access - install the NIKON translation stage
